Do you have any data/experience with using DEF-CS media on Matrigel?

Thin coat matrigel (reduced growth factors) 5µg/cm2 have worked, bit customer needs to optimize. No experience with other matrigel. Stem cell [...]

How long can single cells be cultured in DEF-CS? What's the upper limit of passages caan stem cells survive in DEF-CS without affecting karyotype?

There is no problem to maintain hiPSC in DEF-CS Culture System for more than 20 passages with maintained phenotype and proliferation rate. However, long term culture may affect the genotype and to be on the safe side we recommend around 25 passages in DEF-CS Culture System. We have supporting data on hESC and hope to have similar data on our hiPSC lines within short. Stem cell [...]

Tips for single cell cloning.

If the customer is performing single cell cloning after the electroporation the cells need to recover 5-7 days before starting the single cell cloning. Some points to consider at single cell cloning in addition to the standard DEF-CS protocol: • At single cell cloning (1 well/96-well) DEF-CS GF-3 (diluted 1:1000) should be added at each medium change until the first [...]

Does DEF-CS contain ROCK inhibitors?

Does DEF-CS contain ROCK inhibitors?

No. DEF-CS does not include Rock Inhibitor. We have a different media formulation that when used in combination with our COAT1 reagent results in single cell monolayer culture in the absence of Rock Inhibitor Stem cell [...]
