What kind of pluripotent stem cells can I start with?
The differentiation kits have been shown to work with virtually all of our in-house tested pluripotent stem cell lines (>30 lines). We offer iPS cell lines and ES cell lines as off-the-shelf products, but you can also start with your own pluripotent stem cell lines. Stem Cell [...]
How is the Cellartis iPS Cell to Hepatocyte Differentiation System different from the Cellartis Definitive Endoderm Differentiation Kit with DEF-CS Culture System combined with the Cellartis Hepatocyte Differentiation Kit?
The Cellartis iPS Cell to Hepatocyte Differentiation System includes both the Cellartis Definitive Endoderm Differentiation Kit with DEF-CS Culture Medium and the Cellartis Hepatocyte Differentiation Kit. There are no differences in the composition of the kits if purchased separately or as the complete system. Stem Cell [...]
Does the DEF-CS culture system allow automation?
Yes, the system supports single-cell suspension seeding, allowing control over seeding and harvesting densities and promoting a homogeneous stem cell population. These features, plus no need for cell selection, make the DEF-CS system ideal for mass producing cells with automation. Stem Cell [...]
How many passages can be performed on human iPS cells cultured with the DEF-CS system?
Cells maintain pluripotency and a normal karyotype for at least 25 passages or approximately 65 population doublings. Stem Cell [...]
How frequently should I passage cells grown in DEF-CS
Most human iPS cell lines can be passaged every 3-4 days. We suggest the following weekly schedule: passage on Monday and Thursday, using a slightly higher seeding density on Monday, then change the media on Tuesday, Wednesday, Friday, and Sunday. We recommend seeding 4.0-5.0 x 104 cells/cm2and passaging at a density of 1.5-3.0 x 105 cells/cm2. Stem Cell [...]
