Is the PicoPLEX WGA kit efficient for bacteria?
We are aware that PicoPLEX works with gram-negative bacteria but not sure about gram-positive bacteria - https://www.ncbi.nlm.nih.gov/pmc/articles/PMC4162271/DNA-seq products Care guide Caution [body] This care sheet provides general information only for handling Carolina™ bacterial cultures. When you work with bacteria, it is imperative that you use sterile techniques at all times. Failing to [...]
what is the difference between the PicoPLEX® Single Cell WGA Kit v3 and older generation of PicoPLEX WGA kits?
The PicoPLEX WGA v3 (R300718, R300722, R300723) is a new generation kit and has a significant upgrade in the amplification enzyme, from a standard polymerase to a high-fidelity polymerase. That’s the reason the PicoPLEX® Single Cell WGA Kit v3 kit is recommended mostly for SNV measurements, primarily in oncology; SNP Genotyping. The original PicoPLEX WGA (""version 1"", R30050) or SMARTer [...]
Is it OK to store the library after the Pre-Amplification step with the PicoPLEX kits, prior to the PCR Amplifications step?
The Pre-amplified DNA can be stored at 4 degrees overnight or up to 2 weeks at – 20 degrees. However, it is a good practice to complete the entire protocol including the PCR amplifications step as the DNA yield is extremely low after the Pre-Amplification step and there is a risk of losing some DNA during storage. The amplified WGA is more stable at -20 degrees for several months if [...]
Is PicoPLEX WGA/DNA-seq kit compatible with plant samples such as Pollen?
We have not directly tested the PicoPLEX kits with pollen. But there is a research paper about preparing Illumina libraries from individual pollen nuclei - https://www.frontiersin.org/articles/10.3389/fpls.2017.01620/full#B14 Sequencing of Single Pollen Nuclei Reveals Meiotic Recombination Events at Megabase Resolution and Circumvents Segregation Distortion Caused by Postmeiotic Processes [...]
Can I use first-strand cDNA for the PicoPLEX WGA kit to generate more amplified cDNA?
The PicoPLEX WGA kits cannot be used directly for NGS as the amplified library doesn’t contain primer binding sites for Illumina sequencing. If customers already generated library with the PicoPLEX WGA kit, they can continue using one of the following two approaches - 1. PicoPLEX WGA -> Nextera or Nextera XT or 2. PicoPLEX WGA -> mechanical fragmentation -> ThruPLEX DNA-seq (or any [...]
