The first three nucleotides of the first sequencing read (Read 1) derived from the SMART smRNA Template-Switching Oligo, must be trimmed prior to mapping. 15 As at the 3’-end derived from the 3’ smRNA dT Primer have to be trimmed prior to mapping.
