History of Laminin
1. Laminin are membrane bound and previous purification methods involve harsh conditions.
2. Recombinant laminins can be expressed in mammalian cells and they retain the biological function of natural laminin. During expression, the membrane domains are truncated to ensure soluble expression. The alpha, beta and gamma subunits are cloned separately and co-trransfected into host cell.
3. Laminin E8 was produced by briefly treat the laminin with Elastase. The E8 fragment retains most of the integrin binding activity of Laminin.
4. People start to express the E8 fragments (alpha, beta and gamma) using recombinant technology. The figure below illustrates these pretty nicely. It was found that these recombinant E8 fragments can also support efficient human iPS cell adhesion and expansion.
