How do I analyze double-stranded (ds) cDNA quality?
We recommend analyzing double-stranded (ds) cDNA generated with SMARTer kits using an Agilent 2100 Bioanalyzer and the High Sensitivity DNA Chip (Agilent, Cat. # 5067-4626) before sequencing.Prior to cDNA library analysis, ensure that the electropherogram of the High Sensitivity DNA Ladder is properly displayed: showing a flat baseline, well-resolved ladder peaks, and properly identified Lower and Upper Markers. If the High Sensitivity DNA Ladder electropherogram shows an unexpected pattern, consult the Agilent 2100 Bioanalyzer Expert Maintenance and Troubleshooting Guide.
