What is the difference between ThruPLEX Tag-seq and ThruPLEX Tag-seq HV kits?
The ThruPLEX Tag-seq kits are now considered as legacy kits that are supplied with the TruSeq CD adapters containing the Unique Molecular Tags (UMT). Each UMT is made of a stretch of six degenerate bases, providing more than 4,000 unique sequence combinations. Together, the UMTs on both sides of the fragment produce up to 16 million tags thus ensuring unique labeling of all fragments for identification during data analysis. The ThruPLEX Tag-seq HV kits offer the benefits of the HV kits that allow a higher sample input volume and improved protocols for better coverage of GC-rich templates. Unlike Legacy kits, the ThruPLEX Tag-seq HV kits are supplied with the smaller number of Unique Molecular Identifiers (UMIs known as UMTs in the legacy kits) - 144 UMIs – 12 UMIs in p5 and 12 UMIs in p7 adapters.
